Disease information

Systemic lupus erythematous is an autoimmune disease belonging to the collagenosis group. It is characterized by antibodies against a multitude of body’s organs and organ systems. These antibodies are primarily directed against double stranded DNA (dsDNA). 60 to 80% of the cases of systemic lupus erythematous are positive for these antibodies. However, anti-dsDNA antibodies are also found in association with some other conditions such as autoimmune hepatitis and even in some healthy persons. Anyhow, around 85 percent of the people with these antibodies eventually develop systemic lupus erythematous.

Patients with SLE also tend to have antibodies against nucleosomes which are an exclusive marker for the disease. However, the detection of these requires advanced testing systems and antigens that are free of histone H1, Scl 70 and other non-histone proteins.

Diagnostic Guidelines

Since the disease is based on the production of anti-dsDNA antibodies, the same serve to confirm the diagnosis. Various techniques employed for detection of these antibodies is systemic lupus erythematous are as follows:

  • ELISA
  • EUROLINE
  • EUROASSAY
  • CLIFT

These test differ in their sensitivity and specificity. Of notice is the point that where the conventional CLIFT shows high specificity, IFA CLIFT is a very sensitive test.

Scientists at EUROIMMUN have developed a new test system: the anti-dsDNA-Ncx ELISA which by all standards possess superior diagnostic qualities over all conventional anti-dsDNA ELISA. Our secret lies in using highly purified nucleosomes as the linking substance, which by the virtue of its adhesive qualities binds the antibodies even in low concentrations. This helps in their detection with higher sensitivities.

Supportive Clinical Information

In a comparative clinical study including 378 patients of rheumatic diseases (out of which 209 were those of SLE), our test system yielded an 8% higher sensitivity than conventional anti-dsDNA RIA (Farr Assay), indicating a clear supremacy.

Nevertheless, it’s still better to use different test systems as it helps to identify different subgroups with systemic lupus erythematous.
